"input must be an image stack not a single image" error #7

I expect this tool was extremely beneficial for my research

But i can't use this plugin because "input must be an image stack not a single image" message appears

  • i installed BaSiC via Fiji updater

  • i use fiji 64X version

  • BaSiC is up to date (checked by Fiji updater)

  • image file is Time series stack

  • i upload screenshot about this problem

That image was acquired by Zeiss microscope, so original format was .CZI

Tried BaSiC process which i asked first issue was as follows
import CZI -> "duplicate" single channel with full Time series (yellow) -> "Save as" tiff -> BaSiC

So Technically, i tried BiSiC using single channel image.
i think "c:2/4" on top left derived from original file.
But that doesn't mean it's multichannel.

If there is another way to create "single images (one image per frame)", can you let me know?

Additionally, do you think it is proper to use BiSiC process for FRET ratio images? (For example. Channel 1 intensity / Channel 2 intensity)

I've never used Python before
it will be difficult for me.....

Dear @gyu-ho-choi I think your problem can be solved with the help of ImageJ and/or Fiji alone. There is something in the way how you loaded and/or converted the CZI that makes the BaSiC plugin fail. I think you can get more help about this in the ImageJ and Fiji forums. This should not be too hard to achieve for a person knowledgeable with Fiji.

If you ask in the forums there, can you link it from here, so we can update the solution?
