This wiki contains an overview of Klementine's projects.
Each reaction has its own page, from which you can find an individual issue (corresponding to a notebook page) for each attempt of the reaction. The reaction page also provides the best method and characterisation of the molecule (...so far!).
The sidebar is organised according to target molecules - to find the synthetic steps for a given molecule, click on its name.
To find a specific reaction, look in "Pages", which is sorted alphabetically and also has a search function.
This part of the project focuses on attaching different pendant groups to the cyclam substrate. See this paper for details on previous success with bulky hydrophobic pendant groups.
Previous work in the Rutledge and Todd groups has indicated that "open cyclam" i.e. linear polyamine scaffolds show promising activity against Mtb. There are a number of areas of SAR yet to be explored, summarised in the image below.
So far, only polyamines with naphthyl pendant groups have been tested, and we are interested in trialling alternative bulky, hydrophobic pendant groups. The proposed work involves coupling the azidomethylferrocene and azidoadamantane used in the work above to linear polymine scaffolds.
We are also attempting to explore the effect of chain length by making a series of bis-naphthyl diaminoalkanes that differ only in chain length. Currently, three compounds of this type have been tested, with chain lengths of 2, 4 and 10 carbons. We are focusing on making derivatives with a chain length of 6, 8 and 12 carbons.
